Comparison of the Cytotoxic Effects of Lactobacillus casei Extract with Newcastle Disease Virus in Human Colorectal Cancer

Background and purpose: Colorectal cancer is one of the most prevalent cancers globally, representing a significant public health challenge. Treatment methods include surgery, chemotherapy, and targeted therapies, selected based on disease stage and patient status. Common cancer treatments, such as chemotherapy and radiation therapy, often induce considerable side effects, including fatigue, nausea, hair loss, and blood disorders, which can impact patients' quality of life. Consequently, researchers are focusing on developing novel drugs and targeted therapies aimed at reducing side effects while enhancing treatment efficacy. Innovative therapies, including bacterial and oncolytic viruses, can specifically target cancer cells while sparing healthy tissues. The present study aims to compare the cytotoxic effects of Lactobacillus casei extract with Newcastle disease virus on human colorectal cancer. Materials and methods: Human colorectal cancer cells were cultured and treated with various concentrations of Lactobacillus casei extract and Newcastle disease virus to determine the IC50. Cell viability and apoptosis rates were then assessed using MTT assay and acridine orange/propidium iodide staining. Statistical significance was set at P<0.05 for all evaluations. Results: The results demonstrated that both Lactobacillus casei extract and Newcastle disease virus significantly reduced cell viability and increased apoptosis in a concentration- and time-dependent manner. Caspase-8 and -9 activity measurements indicated that Newcastle disease virus and Lactobacillus casei extract activated both the mitochondrial and extrinsic apoptotic pathways. Conclusion: Based on the findings, it appears that bacterial and oncolytic viruses could be considered as complementary therapeutic options alongside chemotherapy and radiotherapy, following clinical trials.

Gut microbiota – new insight in colorectal cancer pathogenesis

Gut microbiota is a superorganism involved in homeostasis and in pathogenesis. Microbiota composition is influenced by several factors such as: type of birth (cesarean or natural), role of age, role of diet. Pathological consequences of certain type of diet (especially western type of diet) may, in fact, be mediated by gut microbiota alteration. Also few studies have investigated the issue of gut microbiota composition in patients suffering of colorectal cancer, several reports notice relevant differences. Certain pathogenic bacteria in gut microbiota have been extensively studied in relation to their role in colorectal cancer. Thus, Fusobacterium nucleatum, which is abundant in colorectal cancer patient’s colon, may initiate the progression from adenoma to adenocarcinoma. A suggested pattern of oncogenesis in colorectal cancer may be represented by this: dysbiosis-inflammation-oncogenesis. A metanalysis that has been published in 2006 concluded a protective role of probiotics in colorectal cancer and in colonic adenoma.

Manipulation of the gut microbiome may be a promising pathogenic approach in rheumatic inflammatory diseases – A review

A symbiotic association between humans and microbes has been established over time. Disruptions in gut homeostasis have been linked to immune-mediated inflammatory diseases. Factors such as genetics, environment, diet, age, and medication play a major role in shaping the intestinal microbiome. An altered gut microbiome seems to modulate the progression and severity of inflammatory rheumatic diseases. Disruptions in the eubiotic state of the intestinal microbiome might promote intestinal and extraintestinal autoimmune and inflammatory disorders, although the mechanisms involved are not well understood. In this review, we provide a general overview of several studies concerning intestinal microbiome perturbations and their implications in rheumatoid arthritis, ankylosing spondylitis, and systemic lupus erythematosus. Furthermore, we discuss the relationship between diet, intestinal microbiota, and inflammation.

The intestinal microbiome and the role of probiotics/prebiotics in the therapeutic approach of atopic dermatitis: A review

Atopic dermatitis (AD) is the most common chronic inflammatory skin condition, characterized by multiple recurrent eczematous lesions and intense itchiness. It is a multifactorial skin disorder involving an association between genetic and environmental factors that lead to a defect of the epithelial barrier in conjunction with immunological dysregulation. Over the last decade, there has been an increasing understanding of the role of the human microbiota in preserving skin integrity and that a shift in the homeostasis of these microorganisms may lead to dysbiosis and disease. Diversity in the intestinal microbiome and its role in the etiopathogenesis of AD has been described and has become of great interest. In this report, we have reviewed the importance of the gut microbiome and the possible mechanism in the pathogenesis of AD as well as the therapeutic impact of probiotics and prebiotics.

Current Trends, Challenges for Research, and Therapeutic Perspectives in Nutritional Psychiatry

Nutritional interventions in the field of psychiatry have been explored more intensively in the last two decades, but definitive conclusions based on the retrieved data in the literature about the efficacy of this therapeutic approach are difficult to formulate. This observation has multiple explanations, e.g., these therapeutic interventions are very heterogenous, cohort studies usually enroll different psychiatric populations or healthy subjects with minor psychiatric symptoms, nutritional supplements or special diets are commonly recommended as add-ons to the pharmacological interventions (thus making it difficult to differentiate the impact of each therapy on the outcomes), placebo-controlled randomized studies are still very few, etc. Therefore, a narrative review focused on the main challenges reported during the research in the field of nutritional psychiatry (NP) may be useful for clinicians interested in strategies to augment the efficacy of pharmacological treatments or to find new solutions with low risk of adverse events in patients with organic comorbidities or multiple concomitant medications. Data about the efficacy and tolerability of nutritional interventions explored for psychiatric disorders were searched in four electronic databases (PubMed, Cochrane, Clarivate/Web of Science, and EMBASE). Omega-3 polyunsaturated fatty acids supplementation, healthy diet interventions, folate and vitamin B12, S-adenosyl-L-methionine, N-acetyl cysteine, vitamin B1 and B6, vitamin D, minerals, psychobiotics, inositol, and herbal products were the main interventions identified, and their demonstrated efficacy was quite heterogenous, while their tolerability and safety were good. Therefore, NP is a domain of research that may benefit from more good-quality and longer-duration trials, in order to verify the effectiveness of such augmentation strategies to the already validated treatments.
